Open Skype and got to
Tools > Options > General > Audio Settings and make sure your microphone volume is not all the way down. If your microphone is operating properly, the grey bar below the microphone selector wil have green coloring that varies in length based on the volume being detected. If the green doesn't appear, make sure you have the right device selected from the Microphone pull-down. If that doesn't work, try a different microphone. The wires in a microphone are very thin and can break easily. It's not unusual to have a mike work great for quite a while, then suddenly quit.
