Losing settings overnight

I have HP ProLiant ML310 Server running Win Server 2003 however on restart the profiles/preferences go back to default settings. Also, each computer attached to the server lose their settings overnight. These include default printers, toolbars, default paper sizes etc. Each computer defaults back to settings unique to each computer which makes me thinks that this was a local issue, however the server was restarted over weekend and it's toolbar settings went back to default.

Can someone steer me in the right direction? Thanks

What is the user's profile set to in their user account? Is that location available? (I'm assuming that this is a domain).
