Would This Graphics Card Work?

Hello.
I'm upgrading my graphics card on my pci slot, and I was wondering if the ATI 100-436012 Radeon 9250 256MB 128-bit DDR PCI Video Card would be compatible with my eMachines T3958.

Yes that card will work because your motherboard has a PCI Slot and you meet the requirements. But if I were you I would get the AGP 8X Card. Even though your motherboard only supports up to 4X you can still run the card, but it just wont got to 8X it will go to 4X. If you take my advice by getting the AGP slot is universal or it wont work. But Yes the card you suggested works.

with an emachines you will probably have to also upgrade the power supply

It will work but you should go with something much newer. The 9 series is 2004-2005. Unless its just basic pc use.
