Definitely defragment the drive as it will help performance (make sure to do it in Safe Mode for better results).

Check the Properties of the hard drive in the Device Manager (Control Panel -> System -> Hardware tab). Enable write caching if you can and if it's not already enabled, and set the optimization to Optimized for Performance in the Policies tab.

By the way, is it running in DMA or PIO mode?
