to make a selection in PS, press M to activate selection mode. by default it should be in rectangular mode. press shift M to toggle between circular/rectangular modes. when you left click and drag a selection, you can make it concentric/square by holding down shift as you drag.

once you have the selection desired just press ctrl c to copy to the clipboard. the goto file new and select OK. this will automatically create a new document to match the selection you just copied to the clipboard. with the new image open, press crtl v to paste and volia. this new image will now have 2 layers. the one you just pasted and a background layer. you can delete the background if you don't need it. then save the image in a file format that can retain transparency.
